Summary

The purpose of this research is to prepare polyinosinic : polycytidylic acid (poly I : C)microcapsules by intra-liquid desiccation with ethylcellulose (EC) as the polymercoating material. An orthogonal test was used to develop an optimal combination.The optimal combination of encapsulating factors for poly I : C microcapsuleswas found to be as follows: a rotation speed of 700 rpm, 0.4 g magnesiumstearate, 2 % Span-80 in acetone, 2.5 % EC in acetone, and an acetone/paraffinratio of 30:30 (v/v). The pharmaceutical characteristics of size, morphologicalcharacterization, drug loading efficiency, encapsulation yield, and in vitro releaserate were evaluated. The in vitro release rate was directly proportional to theamount of PEG-2000, i. e., with an increase in the amount of PEG-2000,poly I : C release from the microcapsules was more rapid.

Zusammenfassung

Ziel der beschriebenen Forschungsarbeiten ist die Herstellung von poly I:C-Mikrokapseln (Polyinosin-Polycytidylsäure) durch Verkapseln mit Ethylcellulose.Als optimale Bedingungen für die Herstellung wurde mittels Orthogonaltest eineRotationsgeschwindigkeit von 700 rpm bei einem Gehalt von 0,4 g Magnesiumstearat,2 % Span 80 in Aceton, 2,5 % Ethylcellulose in Aceton und ein Aceton/Paraffin-Verhältnis von 30:30 (v/v) ermittelt. Zur pharmazeutischen Charakterisierungwurden die Partikelgröße und deren Morphologie, der Wirkstoffgehalt und dieVerkapselungsrate sowie die In-vitro-Freisetzungsrate untersucht. Die poly I:C-Freisetzungwar dem Gehalt an PEG 2000 direkt proportional; die Freisetzung erfolgtemit zunehmendem PEG 2000-Gehalt rascher.
