Summary
The effect of a polyinosinic:polycytidylic acid copolymer (Poly I:C) on chickembryo fibroblast (CEF) cell viability, anti-Newcastle disease virus (NDV) activityand on IFN- amp;#945; and IFN- amp;#946; levels were determined in vitro. Poly I:C inhibited NDVgrowth in CEF cells in the range of 30–500 amp;#956;g/mL and over 0.75–12 h of treatment.The anti-NDV activity was maximal for 60 amp;#956;g/mL of Poly I:C treatment at12 h, after which it declined. The result of VSV protection assay which was usedto determine IFN-activity showed that Poly I:C inhibited VSV growing on CEF cells,indicating that Poly I:C had immune-stimulating activities. The amount of IFN- amp;#946;in cell supernatant and the transcription levels of IFN- amp;#945; mRNA and IFN- amp;#946; mRNAincreased after Poly I:C treatment, indicating that the anti-viral activity related tothe production of type I IFN. However Poly I:C also showed a toxic effect by inhibitingCEF cell proliferation in a dose-dependent and time-dependent manner. Theresults of this study indicate that the dosage level of Poly I:C must be carefullyconsidered when used in a clinical setting.Zusammenfassung
Die Wirkung eines Polyinosinic:Polycytidylic Säure-Kopolymers (Poly I:C) auf dieCEF-Zelllebensfähigkeit, anti-NDV-Aktivität sowie IFN- amp;#945; und IFN- amp;#946; Level wurde invitro bestimmt. Die Poly I:C hemmte die NDV-Replikation in den CEF-Zellen imBereich von 30-500 ug/mL und bei einer Behandlungsdauer von 0,75–12 Stunden.Die anti-NDV-Aktivität war maximal bei der Behandlungsdauer von Poly I:C60 ug/mL bei 12 Stunden, danach nahm sie ab. Das Ergebnis des VSV ProtectionAssays, der für die Bestimmung der IFN-Aktivitäten eingesetzt wurde, zeigte, dassdie Poly I:C die Replikation des VSVs in den CEF-Zellen unterbinden. Dies deutetdarauf hin, dass Poly I:C eine immun-stimulierende Wirkung hat. Nach der Poly I:C-Behandlung nahm die IFN- amp;#946; Menge im Zellüberstand und der Transkriptionslevelder IFN- amp;#945; mRNA und IFN- amp;#946; mRNA zu. Dies deutet auf den Zusammenhang zwischenanti-viralen Aktivitäten und Produktion des Typ I IFN hin. Allerdings zeigtenPoly I:C auch eine toxische Auswirkung durch Unterbindung der CEF-Zellenproliferationsowohl in einer dosierungs- als auch zeitabhängigen Weise. Das Ergebnisder vorliegenden Untersuchung lässt erkennen, dass bei klinischer Anwendungdie Poly I:C vorsichtig dosiert werden muss.
