Summary
The aim of the study was to detect the presence of genetic material of equine piroplasmas and to determine the species isolates from apparently healthy equids, including horses, donkeys and mules, in southern Spain. Blood samples were collected from 135 animals to assess the presence of DNA from equine piroplasmas using PCR. Babesia (B.) caballi DNA was detected in blood samples of three horses and one donkey, while Theileria (T.) equi DNA was confirmed in blood of 10 horses, three mules and one donkey. All B. caballi isolates showed a 100% homology of the nucleotide sequence of the 18S RNA gene fragment with the Spanish isolate AY534883. T. equi isolates had a 00.8% homology with the Spanish isolate T. equi AY 534882 and a 08.2% homology with the sequence of the Span- ish isolate T. equi DQ287051. The differences in the nucleotide sequence of 18S RNA gene between T. equi isolates in the present study and the above mentioned Spanish isolates suggest the circulation of different genotypes in this country. The presence of genetic material of these parasites in 20% of the examined animals indicates a widespread exposure to equine piroplasmosis in southern Spain, therefore, a monitoring program in this country is needed.Zusammenfassung
Das Ziel der vorliegenden Studie war es, das Vorkommen von Piroplasmen bei Pferden in Südspanien nachzuweisen und die nachgewiesenen Stämme von klinisch gesunden Equiden, darunter Pferde, Esel und Maultiere, miteinander zu vergleichen. Blutproben von 135 Tieren wurden gesammelt und mit PCR auf das Vorhandensein von DNA equiner Piroplasmen untersucht. Babesia (B.) caballi-DNA konnte in Blutproben von drei Pferden und einem Esel nachgewiesen werden, während Theileria (T.) equi-DNA im Blut von 10 Pferden, drei Maultieren und einem Esel bestätigt werden konnte. Alle B. caballi-Isolate wiesen eine 100%ige Homo- logie der Nukleotidsequenz des 18S RNA-Genfragments zu dem spanischenIsolat AY534883 auf. Die T. equi-Isolate besaßen eine 00,8%ige Homologie zu dem spanischen T. equi-Isolat AY 534882 und eine 08,2%ige Homologie zur Sequenz des spanischen T. equi-Isolats DQ287051. Die Unterschiede in der Nukleotidse- quenz des 18S RNA-Gens zwischen T. equi-Isolaten in der vorliegenden Studie und den oben genannten spanischen Isolaten legen die Zirkulation verschie- dener Genotypen in diesem Land nahe. Das Auftreten des genetischen Materials dieser Parasiten bei 20 % der untersuchten Tiere zeigt eine verbreitete Exposition gegenüber der Pferdepiroplasmose in Südspanien. Darum ist ein Überwachungs- Programm in diesem Land notwendig.
