Zusammenfassung
Das feline Fibrosarkom ist ein spontan auftretender,maligner Bindegewebstumor, der mit einer Rezidivratevon etwa 70 %, auch nach radikaler Tumorexstirpation,eine sehr schlechte Prognose hat. Adjuvant zur Operationwurde bereits in verschiedenen klinischen Studien eineZytokin-Gentherapie eingesetzt. Im Rahmen dieses Projektswurde ein nonvirales Gentherapeutikum entwickelt. Diesesbesteht aus einem resorbierbaren Kollagenschwamm,der mit Plasmiden in einer dauerstabilen Formulierungbeschichtet ist. Die eingesetzten Plasmide kodieren für diefelinen Zytokine Interleukin-2 (IL-2), Interferon amp;#947; (IFN amp;#947;) undGranulozyten-Makrophagen-Kolonie-stimulierender-Faktor(GM-CSF). Die Bildung einer Antitumorimmunität soll einlokales Rezidiv des Fibrosarkoms verhindern oder die rezidivfreieZeit der Katzen verlängern.Die für felines (fe)IL-2, feIFN amp;#947; und feGM-CSF kodierendencDNAs werden aus mononukleären peripheren Blutzellenmittels PCR kloniert und entsprechen den publiziertenSequenzen. Die rekombinanten Proteine werden in einerSäugerzelllinie exprimiert. In Proliferationsassays kann dieBioaktivität von feIL-2 und feGM-CSF demonstriert werden.Die biologische Aktivität von feIFN amp;#947; wird durchflusszytometrischmit MHC-I- und -II-Induktionstests auf felinenFibrosarkom-Zellen nachgewiesen. Nach Komplexierungder Plasmid-DNA mit dem Polykation Polyethyleniminwird dieser Komplex mit protektiven Kopolymeren mittelsLyophilisation auf einen resorbierbaren Kollagenschwammaufgebracht. Dieser beschichtete Schwamm kann bis zurImplantation ins Tumorbett steril bei 4 °C gelagert werden.Ziel ist die Entwicklung einer adjuvanten Immuntherapiezur derzeitigen Standardtherapie des felinen Fibrosarkomsund damit zunächst der Einsatz dieses Gentherapeutikumsin einer Phase-I-Studie.Summary
Cloning of the feline cytokines IL-2, IFN amp;#947; and GM-CSF for anadjuvant nonviral gene therapy of feline fibrosarcomaThe feline fibrosarcoma is a spontaneously occurringmalignant soft tissue sarcoma. Due to its high recurrencerate of approximately 70%, the prognosis is very poor evenafter radical surgical excision. So far different cytokinegene therapy protocols have been carried out adjuvant tosurgical tumour excision. A nonviral gene delivery systemhas been developed in this project. A collagen spongewas loaded with plasmids in a copolymer-protected genevector (COPROG) formulation. The plasmids were codingfor interleukin-2 (feIL-2), interferon amp;#947; (feIFN amp;#947;) and granulocyte-macrophage colony-stimulating factor (feGM-CSF).The aim of this gene therapeutic approach is to enhanceanti-tumour immunity, extend the tumour-free survivaltime or to decrease the recurrence rate in fibrosarcomaaffectedcats. Indeed, the final aim is the development ofan immunotherapy adjuvant to the surgical excision of thefeline fibrosarcoma. The initial goal reported in this paper,however, was to clone these three cytokines as well as toprove the biological activity of the recombinant proteins.The cDNA coding for feIL-2, feIFN amp;#947; and feGM-CSF wascloned via PCR using feline peripheral mononuclear bloodcells. The recombinant proteins were expressed in a mammaliancell line. The feIL-2 and feGM-CSF bioactivity wasdemonstrated in proliferation assays. The biological activityof feIFN amp;#947; was measured by FACS analysis. Feline IFN amp;#947; inducesan increased MHC I and II expression on feline fibrosarcomacells. After plasmid complexation with the polycationpolyethylenimine and protective copolymers, the collagensponge was soaked with the gene vectors and lyophilised.It was stored under sterile conditions until implantation inthe tumour bed of fibrosarcoma-affected cats. A phase Idose escalation study was conducted using the vectorloadedcollagen sponge.
