Etablierung einer standardisierten und optimierten regiospezifischen intranasalen Applikationsmethode an der Maus mit Hilfe von 3D-Modellen unter 3R-Kriterien

Summary
The aim of the „3D for 3R“ project was to generate anatomical 3D models of the murine nasal cavity with rapid prototyping to establish a refined region-specific intranasal drug delivery technique targeting either the respiratory or the olfactory epithelium in mice. The anatomical structure of the murine nasal cavity was resolved by x-ray computer tomography and the data sets of the CT-scans were used to construct anatomical prototypes that were manufactured by 3D-printing, stereolithography or vacuum moulding. In the present study we show the successful construction of such anatomical 3D models and their use to establish intranasal application techniques under 3R criteria to minimize the use of laboratory animals.
Zusammenfassung
Das Ziel des Projekts „3D für 3R“ war die Erstellung von anatomischen 3D-Modellen der Nasenhöhle von Mäusen, um eine verfeinerte regio-spezifische intranasale Darreichungsmethode zu entwickeln, die spezifisch entweder das respiratorische oder das olfaktorische Epithel erreichen soll. Die Anatomie der murinen Nasenhöhle wurde durch Computertomographie dargestellt und die Datensätze wurden verwendet, um anatomisch korrekte Modelle zu konstruieren, die mittels 3D-Druck, Stereolithographie oder Vakuumgussverfahren hergestellt wurden. In der vorliegenden Studie zeigen wir die erfolgreiche Konstruktion solcher anatomischer 3D-Modelle und deren Verwendung zur Etablierung intranasaler Applikationstechniken unter 3R-Kriterien, um den Einsatz von Labortieren zu minimieren.
