Amputation and regeneration of limbs in the Axolotl (Ambystomamexicanum)

Zusammenfassung
Die biologischen Prozesse, mit denen Lebewesen beschädigtes Gewebe heilen oder regenerieren, beinhalten komplexe biochemische und zelluläre Vorgänge. In einigenSpezies kommt es dabei zu beachtlichen Regenerationsleistungen. Dazu gehört die Gliedmaßenregeneration der Urodelen (Schwanzlurche). Eine wichtige Voraussetzung
zum Ersatz des fehlenden Gliedes ist hier eine lokale Antwort des Stumpfgewebes. Aus diesem Grund darf nach einer Gliedmaßenamputation beim Axolotl das Wundgewebe
nicht mittels Wundnaht verschlossen werden, weil dadurch der auslösende Reiz zur Ausbildung des Wundepithels unterbunden wird. Unter Umständen ist zur Optimierung
des Heilungsverlaufes eine Amputation im gesunden Gewebe notwendig. Eine geeignete Narkose bei Eingriffen an innerviertem Gewebe ist ein Tauchbad mit MS-222
(Ethyl-3-Aminobenzoat-Methansulfonat, Finquel®; Argent Chemical Laboratories, USA). Eine Lokalanästhesie sowie eine Desinfektion des Gewebes sind nicht angezeigt, da
hierdurch Gewebsnekrosen hervorgerufen werden können.
Summary
Biological processes by which living creatures heal or regenerate damaged tissues comprise complex biochemical and cellular processes. In some species, remarkable results in regeneration are accomplished. One example is the limb regeneration of urodeles, and in this particular case, the axolotl. An important prerequisite for the replacementof a lost limb is a local reaction of the stump tissue. For this reason, after a limb amputation, the wound tissue must not be closed by sutures, by which the inductive
stimulus to build up wound epithelium is suppressed. In certain cases, amputation in the healthy tissue may be required for optimisation of the healing process. Appropriate
anaesthesia when performing surgery in enervated tissue in these animals is the immersion into MS 222 (ethyl 3-aminobenzoate methanesulphonic acid; Finquel®, Argent,
USA). Disinfection and local anaesthesia are not indicated because they might provoke tissue necrosis.
