Summary:<\/span>
The aim of the investigation was to compare four published PCR methods for specific detection of Pasteurella multocida<\/span>. 85 strains of P. multocida<\/span> and 13 strains of other taxa were included in the comparison of four published PCR methods based on kmt1<\/span>, 23S rRNA and transcriptional regulator genes pm0762 and pm1231 as targets, respectively. Sensitivities were calculated to 100 and 80% for the kmt1 and 23S rRNA based PCRs, respectively and specificities to 92 and 23%, respectively of the kmt1<\/span> and 23S rRNA based PCRs. The methods based on transcriptional regulator genes tested two Bisgaard taxon 45 strains positive in addition to one Bisgaard taxon 16 strain. Only the kmt1 <\/span>method detected P. multocida<\/span> with the expected sensitivity and specificity and this method is recommended for identification of the species.

Das Ziel der Untersuchung war, vier publizierte PCR-Verfahren auf ihre Eignung zur Differenzierung von Pasteurella multocida<\/span> zu vergleichen. 85 St\u00e4mme von
P. multocida<\/span> und 13 St\u00e4mme anderer Spezies wurden f\u00fcr den Vergleich der PCR-Methoden kmt1<\/span>, 23S rRNA, Transkriptions-regulierende Gene pm0762 und pm1231 herangezogen. Die Sensitivit\u00e4t wurde mit 100 und 80 % f\u00fcr kmt1 und die 23S rRNA basierten PCR-Methoden bestimmt. Dem gegen\u00fcber lagen die Spezifit\u00e4ten bei 92 und 23 % f\u00fcr kmt1 <\/span>und die 23s rRNA basierten PCR-Methoden. Die 23S rRNA basierten PCR-Methoden testeten zwei Bisgaard Taxon 45 St\u00e4mme und ein Bisgaard Taxon 16 Stamm positiv und wurden daher als ungeeignet verworfen.
